故事舞台搬到了人類發源地非洲，在這塊炎熱的大地上擁有並非殭屍也不屬於寄生獸的敵人
飽經歷練的克里斯（Chris） 將與新搭檔席娃（Sheva）共同面對潛藏於黑暗大陸的神祕生化危機。
登場敵人部分 除了有玩家所熟悉的生化殭屍犬，貌似巨大蝙蝠的全新登場生化合成怪獸 以及受到寄生而失去理智成為嗜血殺人狂的電鋸人、巨斧劊子手等也都將一一登場。
在夥伴合作模式方面，克里斯與席娃將以火力互相掩護，彌補各自的視線死角，並視情況給予夥伴必要的協助還可以透過共同行動來攀高越牆，克服地形障礙。在 PC 版當中，特別新增新傭兵模式，敵人將比遊樂器版本多了三倍 同時克里斯與席娃都添置了新服裝，席娃將穿著美麗的秘書裝帥氣地殺出生路。
